Larazotide

A peptide studied for tightening the gut barrier, researched mainly in celiac disease — one of the more clinically studied gut peptides.

In plain terms: Larazotide is a peptide studied for tightening up the "seams" between cells in the gut lining, so less leaks through. Most of the research is in celiac disease, and it has been tested fairly seriously in people — though it is not approved.

What it is

Larazotide is a lab-made peptide studied for its effect on the gut barrier — the lining that controls what passes from the intestine into the body. It is designed to act only inside the gut rather than being absorbed.

It has one of the more substantial human research records among gut peptides, mostly in celiac disease.

Mechanism

The gut lining has microscopic "seams" between its cells called tight junctions. When these loosen, more can pass through (sometimes called a "leaky" gut). Larazotide is studied for helping keep these seams tight, reducing what gets through.

This has been studied in cell, animal, and human research focused on celiac disease.

Research history

Larazotide came out of research into the gut barrier and the protein zonulin. It progressed through human clinical trials for celiac disease in the 2010s, with results in major gastroenterology journals.

State of the literature

Larazotide has been studied in registered human trials for celiac disease, with mixed late-stage results. It is not an approved drug. Research continues.

What to keep in mind

Larazotide has more human data than most gut peptides, but its late-stage celiac trials did not clearly succeed — a reminder that even well-studied compounds often do not pan out. "Studied for leaky gut" is also a phrase marketing loves; the actual research is specific to celiac disease, not a general gut fix. Read the trial results, not the slogans.
